Interests: The overall goal of my research is to investigate the epigenetic mechanisms that are involved in the normal and malignant hematopoiesis to further delineate how these networks are regulated in acute myeloid leukemia (AML). I have also a broad background in biochemistry and molecular biology with specific training and expertise on applying molecular biological and bioinformatics technologies to studying the mechanism of chromatin organization and remodeling regulation in human normal and malignant hematopoiesis. One of my research projects published in Blood, I have found one of critical CTCF boundary between HOXA7/HOXA9 genes (CBS7/9) remodels chromatin domain and induces aberrant HOX gene expression in acute myeloid leukemia. In order to figure out whether this critical boundary CBS7/9 mediated chromatin structure organization for leukemogenesis in vitro and in vivo, 4C-seq, ATAC-seq and HiC-seq have been applied to clarify this mechanism. As an assistant professor in Penn State University, I are focused on the bioinformatics analysis and have a good collaboration with others to analyze Next Generation Sequencing data to figure out the role of HOX associated lncRNAs (long non-coding RNAs) in leukemogenesis. HOTTIP as one of critical lncRNAs in HOXA loci, which has a coordinate function with CTCF boundary to control the HOXA chromatin structure organization and HOXA genes transcription, and lots of next generation sequence (NGS) datasets from patient tumor samples and cancer cell lines, such as ATAC-seq, RNA-seq, 4C-seq, ChIP-seq and Hi-C-seq, have been used to elucidate this hypothesis. Thus, combination of molecular biological and bioinformatics technologies to investigate the mechanism of human normal and malignant hematopoiesis will well support in my future research projects
